In vitro study | Z-VAD-FMK(10 mM) acted on THP.1 cells to inhibit apoptosis. Z-VAD-FMK(10 μm) acts on THP.1 cell lysate to inhibit activation of PARP protease activity. Z-VAD-FMK(10 mM) inhibits CPP32 processing in intact THP.1 and Jurkat cells. Z-VAD-FMK(50 μm) co-treatment of HL60 cells with Camptothecin abolished the apoptotic morphology of Camptothecin-treated cells. Z-VAD-FMK(50 μm) inhibits Camptothecin-induced DNA division in HL60 cells. Z-VAD-FMK (50 μm) increased the survival percentage of transfected cells from 26% to 63% in S2 cells. In vivo study | Studies have shown that the administration of Z-VAD-FMK in vivo is not toxic and can inhibit apoptosis in animal models. In mice, intraperitoneal injection of HK-GBS can cause preterm labor, while pretreatment with Z-VAD-FMK can delay the phenomenon of preterm labor. In OVA-sensitive mice, z-VAD-fmk treatment inhibited allergen-induced leukocyte infiltration. Systemic injection of z-VAD-fmk prior to OVA sensitization reduced the accumulation of inflammatory cells, mucus hypersecretion, and Th2 cytokine release in OVA-sensitive/challenged mice.
